ABSTRACT

Multiple morphological abnormalities of the sperm flagella (MMAF) is a severe form of asthenozoospermia categorized by immotile spermatozoa with abnormal flagella in ejaculate. Whole-exome sequencing (WES) is used to detect pathogenic variants in patients with MMAF. In this study, a novel homozygous frameshift variant (c.6158_6159insT) in dynein axonemal heavy chain 8 (DNAH8) from two infertile brothers with MMAF in a consanguineous Pakistani family was identified by WES. Reverse transcription-polymerase chain reaction (RT-PCR) confirmed DNAH8 mRNA decay in these patients with the DNAH8 mutation. Hematoxylin-eosin staining and transmission electron microscopy revealed highly divergent morphology and ultrastructure of sperm flagella in these patients. Furthermore, an immunofluorescence assay showed the absence of DNAH8 and a reduction in its associated protein DNAH17 in the patients' spermatozoa. Collectively, our study expands the phenotypic spectrum of patients with DNAH8-related MMAF worldwide.

ABSTRACT

Objective: To understand the impact of platelet associated immunoglobulin G (PAIgG)/platelet associated immunoglobulin M (PAIgM) on severity of dengue virus infection leading to thrombocytopenia. Methods: In this study we examined a total of 52 patients who were having secondary infection of dengue in acute phase by using competitive ELISA. Results: A decrease in the platelet count was observed at the acute phase of infection while all along the recovery stage the count of platelet was significantly increased. A significant decrease was observed in PAIgG and PAIgM in these subjects. Inverse correlation was found between platelets count and PAIgG/PAIgM among the subjects studied. In the platelets elution from ten subjects, anti-dengue virus immunoglobulin G and immunoglobulin M were observed. PAIgG and PAIgM with inclined levels were higher in dengue hemorrhagic fever than the classical dengue fever. In the development of dengue hemorrhagic fever PAIgM inclined level was independently associated with high specificity, showing a possible indication of dengue hemorrhagic fever. Conclusions: This study suggests that in secondary dengue virus infection, the PAIgG and PAIgM levels, and the activity of anti-dengue virus play key roles, both in the development and severity of the disease.

ABSTRACT

Urinary incontinence can be distressful and may affect the quality of life to a great extent. Common types of UI are stress incontinence, urge incontinence and mixed incontinence. UI is more prevalent in older people and more frequently diagnosed in females than males. Efforts are made to investigate the primary disorder which causes incontinence, as UI is mostly secondary to an underlying medical condition. This study included patients diagnosed with UI and was conducted during one year period extending from February 2013 to January 2014. The data also constituted of a self-administered questionnaire to access the patient's knowledge regarding their disease. Ethical consent was obtained from the hospital where data were collected and oral and written consents were taken from all the participants prior to their participation. The study included 332 patients with a response rate of 73.7%. Overall, 254 males and 278 females participated. 64.9% of the male participants had urge incontinence followed by 26.6% having stress incontinence, while only 8.44% of the males were diagnosed with mixed incontinence. In females 77.2% had stress incontinence followed by urge incontinence [15.1%] and only 7.3% had Mixed urinary incontinence. 24% of males and 44.3% of females believed that pelvic exercises can prevent or treat UI. 34.6% of the total participants [40% females,27.9% males] had an understanding that weak anatomy contributes to the development of UI. Stress incontinence is more common in women, while the dominant type in men is urge continence. All patients, especially pregnant women should be provided information regarding UI in order to efficiently cope up with the condition if encountered

ABSTRACT

Blood borne diseases impose heavy burdens on national economies and individual families due to costs arising from acute and chronic morbidity and mortality. Globally, 2 billion people are infected with hepatitis B virus [HBV]. An estimated 170 million persons are chronically infected with hepatitis C virus [HCV] and 3-4 million persons are newly infected each year. Transmission of hepatitis B and C through infected needles, blades, etc. is well known. Barbers are the important parts of community, as almost every man needs at least monthly visit to barber for hair dressing or shaving. In Pakistan the prevalence of shaving by barbers is reported to be as high as 34%-49% of the male population and it is important to know how barbers perceive the risks in relation to prevention of transmission of hepatitis B and C. To assess the knowledge and attitude of barbers regarding HBV and HCV and find out practices among barbers which may be responsible for transmission of HBV and HCV. Cross-Sectional Study. This study was carried out in the Larkana city during the months of March 2010 to April 2012. A list of the barbers was obtained from the union of barbers and sample was drawn from that sampling frame. There were 422 barbers shops in the city. For sampling purpose Larkana city was arbitrarily divided into five zones [East, West, Central, South and North] and then 10 shops randomly selected from each zone. There were 156 barbers available and eligible for interview. After informed verbal consent, 132 barbers agreed to participate in the study. Data was entered, analyzed by Statistical Program for Social Sciences [SPSS] version 13. A total 132 barbers were interviewed. The mean age of respondents was 28 +/- 10 years, mean years of experience in barbering was 12.7 +/- 10. More than half [63.6%] respondents were married. Majority of the respondents [84.8%] were resident of urban areas. 64% barbers have had knowledge that hepatitis B and C can be transmitted through blood transfusion and sexual contact. Only 6.1% had been vaccinated against the hepatitis B. 70% of the barbers disposed of used blades in regular garbage. 82% washed hands before and after shaving each client, 92.4% cleaned their instruments with disinfectant after shaving the, 99.2% barbers change the blade for each client. The knowledge of barbers regarding the transmission of hepatitis B and C was fairly good but majority of the barbers were not vaccinated against Hepatitis B though they were knew that vaccine is available against the hepatitis B

ABSTRACT

To see the gross qualitative parameters such as general appearance of rats and their testes after long and short term use of Sildenafil Citrate. As very little attention has been given to explore the effects of a sildenafil citrate on histological aspects of testes, hence this experimental study was designed to check whether the drug which is being used indiscriminately in our country and abroad is safe or it has any harmful effect on the architecture of rat testis. Experimental Study. This study was conducted in departments of Anatomy and Histopathology Shaikh Zayed Hospital Lahore for a period of six weeks from 02.05.2008 to 17.06.2008. Sample size consisted of 45 animals, divided into Group A [Control], Group B and C [Experimental], Each group was consisting of 15 animals. Physical examination of rats and their testis was done every day by the author himself and recorded. After giving drug, on inspection of rats all the animals were active and healthy and the gross appearance of the testes was normal except in an animal, in which the testes were smaller than the associates of the same group. Eating habits of all the animals were normal, taking food and water freely. After half an hour of giving the drug, they were mounting over each other and looked aggressive. The comparison of all the groups, A vs B, A vs C and B vs C remained statistically non -significant [P>0.05].
